Project Overview

In bioarchaeology, sequencing DNA from ancient organisms is a crucial source of information. However, such DNA is highly fragmented and single-stranded. Therefore, these DNAs are not compatible with commonly used protocols for sequencing library preparation, as these methods are typically adapted only for double-stranded DNA. To resolve this problem, we developed a single-strand DNA (ssDNA) compatible sequencing library preparation technique, SDL-TOPO, and have optimized its use for DNA obtained from ancient organisms. In this research project, we will further optimize and improve the SDL-TOPO, making it available to those who wish to utilize it. Additionally, we will collaboratively address technical challenges faced by researchers in this field from the perspectives of biotechnology and genome technology.
